The aim of this study was to evaluate an osteopromotive technique, using a bioabsorbable membrane... more The aim of this study was to evaluate an osteopromotive technique, using a bioabsorbable membrane, for its ability to restitute bone over buccal fenestration and dehiscence defects following fixture installation. 11 patients requiring dental implant treatment and exhibiting sufficient vertical height of the maxilla and compromised bucco-palatal dimensions, as determined clinically and radiographically, were included in the study. 17 Brinemark@ titanium fixtures were placed with buccal defects which were augmented by a bioabsorbable membrane, Resolut@. No complications were observed postoperatively. At 6-8 months, abutment connection was performed, and clinical evaluation of the healed defect area was made. The number of exposed buccal threads at fixture installation (median 8; range 2-19), and abutment connection (median 0; range 0-5), respectively, was compared. Out of the 17 fixtures; 14 exhibited complete coverage with bone, whereas 3 showed some remaining threads. A small punch biopsy taken at abutment connection in an area where the membrane had been placed showed a combination of dense connective tissue and bone. Radiographic evaluation of the marginal periimplant bone level is in progress and results to date show a median bone loss of 1.2 mm after a loading period of 4-6 months. Results show that fixture dehiscence and fenestrations, augmented with this bioabsorbable membrane, demonstrate a highly significant amount of new bone formation.

In rodents, Homer proteins and mRNAs have been shown to be expressed in various postnatal tissues and to be enriched in brain. However, whether the Homers are expressed in developing tissues is hitherto largely unknown. In this work, we used immunohistochemistry and in situ hybridization to analyze the expression patterns of Homer1, Homer2 and Homer3 in developing cephalic structures. Our study revealed that the three Homer proteins and their encoding genes are expressed in a wide range of developing tissues and organs, including the brain, tooth, eye, cochlea, salivary glands, olfactory and respiratory mucosae, bone and taste buds. In several embryonic structures, components of the Hedgehog and retinoic acid pathways, two potent players in development and disease are expressed and operate in the same or adjacent tissues and cells. Yet whether and, if so, how these pathways interact during organogenesis is, to a large extent, unclear. The interaction between signaling pathways is a central question in the study of organogenesis. U... more The interaction between signaling pathways is a central question in the study of organogenesis. Using the developing murine tongue as a model, we uncovered unknown relationships between Sonic hedgehog (SHH) and retinoic acid (RA) signaling. Genetic loss of SHH signaling leads to enhanced RA activity subsequent to loss of SHH-dependent expression of Cyp26a1 and Cyp26c1. This causes a cell identity switch, prompting the epithelium of the tongue to form heterotopic minor salivary glands and to overproduce oversized taste buds. At developmental stages during which Wnt10b expression normally ceases and Shh becomes confined to taste bud cells, loss of SHH inputs causes the lingual epithelium to undergo an ectopic and anachronic expression of Shh and Wnt10b in the basal layer, specifying de novo taste placode induction. Surprisingly, in the absence of SHH signaling, lingual epithelial cells adopted a Merkel cell fate, but this was not caused by enhanced RA signaling. We show that RA promotes, whereas SHH, acting strictly within the lingual epithelium, inhibits taste placode and lingual gland formation by thwarting RA activity. These findings reveal key functions for SHH and RA in cell fate specification in the lingual epithelium and aid in deciphering the molecular mechanisms that assign cell identity.

By using a mechanical hindrance, a porous expanded polytetrafluoroethylene (e-PTFE) membrane, it is possible to prevent fibroblasts and other soft connective tissue cells from entering the defect, thereby allowing osteogenesis to occur unhindered. As evidenced in several investigations, this osteopromotive membrane technique causes a strongly improved bone regeneration of well defined osseous lesions. Hyperbaric oxygen treatment has also been shown to accelerate bone healing. In this study the value of combining the two techniques was investigated. Dentin may be regarded as a calcified connective tissue. In this respect, as well as in its mode of formation, it is closely related to bone. Using dentinogenesis as an experimental model to study biomineralization provides several practical advantages, and the results may be extrapolated to understand similar processes in other tissues, primarily bone. After describing dentin structure and composition, this review discusses items such as the morphology of dentinogenesis; the dentinogenically active odontoblast, transport, and concentrations of mineral ions; the constituents of the dentin organic matrix; and the presumed mechanisms involved in mineral formation.

After 12 weeks, the sites revealed a massive ingrowth of soft connective tissue, forming a transosseous core filling the defects. Upon reentry, the soft tissue inside the remaining bone defects was removed. On one side of the jaw, the defect was covered both buccally and lingually with an expanded polytetrafluoroethylene (e-PTFE) membrane, but on the other side no membrane was placed. Histologic analysis after 6 weeks revealed an essentially complete healing with bone of the membrane-covered defects. No cartilage was present in any of the specimens. At the control sites (no membrane), the amount of newly produced bone showed variations, most through defects revealing the presence of a remaining central portion of connective tissue. This report describes findings in microbial samples from 271 patients with lesions of the oral mu... more This report describes findings in microbial samples from 271 patients with lesions of the oral mucous membranes. Analyses were made on the basis of direct microscopy and cultivation on nonselective and selective media incubated under aerobic and anaerobic conditions. Growth of bacteria was semiquantitatively determined according to a five-grade scale. Among the 134 samples for direct microscopy, fungi in yeast as well as hyphae phase, were identified in 39 cases. Among the cultivated samples, Candida albicans was detected in 107 cases, of which 71 were full-denture wearers. Staphylococcus aureus, coliform bacteria and Klebsiella were the most frequently isolated "nonoral" bacteria. In some cases also Streptococcus pyogenes, Haemophilus influenzae, Pseudomonas or other gram-negative aerobic bacteria were found. In 84 cases the bacteria mentioned were registered as single "nonoral" infection, and in 63 cases combinations of "nonoral" species occurred. In ...
The non-collagenous proteins of rat dentin that remain firmly bound to the matrix after demineral... more The non-collagenous proteins of rat dentin that remain firmly bound to the matrix after demineralization were studied in order to ascertain if they are covalently linked to insoluble dentin collagen. After solubilization with CNBr or with bacterial collagenase, unusually small amounts of dentin phosphoprotein were detected in the matrix. The phosphoprotein obtained by CNBr digestion of the matrix was separated from collagen peptides using two chromatographic steps. Thus even this small quantity of phosphoprotein found in decalcified rat dentin matrix was not covalently bound to collagen.

For screening tests of bacterial beta-glucuronidase (EC 3.2.1.31) activity an agar plate (MUG pla... more For screening tests of bacterial beta-glucuronidase (EC 3.2.1.31) activity an agar plate (MUG plate) containing the enzyme substrate, 4-methylumbelliferyl-beta-D-glucuronide, was developed. In the presence of beta-glucuronidase, the strongly fluorescent 4-methylumbelliferone, which can be detected by ultraviolet light, was liberated. For tests of fastidious bacteria, the MUG-plate was enriched with supplements containing heat-labile growth factors without influencing the reaction. The plate was found to be suitable for screening tests. The sensitivity of the MUG-plate was determined by means of fluorimetric method. With the present method, beta-glucuronidase activities were demonstrated in some well-known beta-glucuronidae-producing microorganisms as well as in Bacteroides and Corynebacterium spp.

The synthesis of glycosaminoglycans (GAGs), the carboxyhydrate side chains of proteoglycans, in tissue from the lower uterine segment of term pregnant women was investigated in vitro by measurement of 35SO4 and [14C]glucosamine incorporation. Prostaglandin E2 and oestradiol-17 beta significantly increased the synthesis of sulphated GAGs but decreased the incorporation of [14C]glucosamine, while relaxin, prostaglandin F2 alpha and oxytocin had no significant effect. In the present study, a new membrane technique has been utilized in an attempt to improve bone healing. By means of an inert, porous membrane, placed in close contact with the bone surface, a secluded space is created which can only be repopulated by cells from the adjacent bone. Thus, osteogenesis is able to occur without interference from other tissue types. Through-and-through bone defects were produced bilaterally (1) in edentulous areas of monkey (n = 5) mandibles, and (2) in conjunction with apicectomy of the lateral maxillary incisors, also in monkeys (n = 7). The aim of the present study was to determine whether bone quality, as assessed by osteometry and selected haematologic and urinary tests, influences the integration of implants, and whether such data can be prognostically useful. Relative bone mass density (BMD%) differed significantly among these patients as compared to age- and sex-matched control patients receiving the same reconstructive treatment (P=0.01). Other parameters tested did not demonstrate any significant differences. In addition to local complications, general disorders, such as osteoporosis, must be considered in cases of excessive implant loss.

The non-collagenous proteins of rat dentin that remain firmly bound to the matrix after demineral... more The non-collagenous proteins of rat dentin that remain firmly bound to the matrix after demineralization were studied in order to ascertain if they are covalently linked to insoluble dentin collagen. After solubilization with CNBr or with bacterial collagenase, unusually small amounts of dentin phosphoprotein were detected in the matrix. The phosphoprotein obtained by CNBr digestion of the matrix was separated from collagen peptides using two chromatographic steps. Thus even this small quantity of phosphoprotein found in decalcified rat dentin matrix was not covalently bound to collagen.

For screening tests of bacterial beta-glucuronidase (EC 3.2.1.31) activity an agar plate (MUG pla... more For screening tests of bacterial beta-glucuronidase (EC 3.2.1.31) activity an agar plate (MUG plate) containing the enzyme substrate, 4-methylumbelliferyl-beta-D-glucuronide, was developed. In the presence of beta-glucuronidase, the strongly fluorescent 4-methylumbelliferone, which can be detected by ultraviolet light, was liberated. For tests of fastidious bacteria, the MUG-plate was enriched with supplements containing heat-labile growth factors without influencing the reaction. The plate was found to be suitable for screening tests. The sensitivity of the MUG-plate was determined by means of fluorimetric method. With the present method, beta-glucuronidase activities were demonstrated in some well-known beta-glucuronidae-producing microorganisms as well as in Bacteroides and Corynebacterium spp.

Proteoglycans, being a main constituent of the extracellular matrix, influence the physical properties of the tissue and play an important regulatory role for a number of functional events. The synthesis of glycosaminoglycans (GAGs), the carboxyhydrate side chains of proteoglycans, in tissue from the lower uterine segment of term pregnant women was investigated in vitro by measurement of 35SO4 and [14C]glucosamine incorporation. Prostaglandin E2 and oestradiol-17 beta significantly increased the synthesis of sulphated GAGs but decreased the incorporation of [14C]glucosamine, while relaxin, prostaglandin F2 alpha and oxytocin had no significant effect. In the present study, a new membrane technique has been utilized in an attempt to improve bone healing. By means of an inert, porous membrane, placed in close contact with the bone surface, a secluded space is created which can only be repopulated by cells from the adjacent bone. Thus, osteogenesis is able to occur without interference from other tissue types. Through-and-through bone defects were produced bilaterally (1) in edentulous areas of monkey (n = 5) mandibles, and (2) in conjunction with apicectomy of the lateral maxillary incisors, also in monkeys (n = 7). On one side, the defects were covered buccally as well as lingually/palatally with expanded PTFE membranes, whereas the defects on the other side served as controls (no membrane). In a retrospective analysis of 49 patients who received bone graft augmentation to the maxillary ... more In a retrospective analysis of 49 patients who received bone graft augmentation to the maxillary sinuses in conjunction with implant insertion, 11 patients had a significantly reduced success rate. The aim of the present study was to determine whether bone quality, as assessed by osteometry and selected haematologic and urinary tests, influences the integration of implants, and whether such data can be prognostically useful. Relative bone mass density (BMD%) differed significantly among these patients as compared to age- and sex-matched control patients receiving the same reconstructive treatment (P=0.01). Other parameters tested did not demonstrate any significant differences. In addition to local complications, general disorders, such as osteoporosis, must be considered in cases of excessive implant loss.

The Saints began its existence as a serene African-American spiritual, presumably well spread in the American South. Initially it was recorded by jubilee singers, self-accompanied vocalists and country musicians in the 1920:ies. The tune later became the jazz standard, worn out to disgrace. It was eventually taken up by a plethora of rock singers, gospel artists, folk singers, comedians, dance orchestras and marching bands, and the song also made its way into children's music. The Saints has become emblematic of the city of New Orleans, LA and even provided the name to its American football team. All the time, however, the tune has maintained its role as a religious hymn, sung by individuals, in congregations and played at funerals.
