Čeština: Dvě semínka vápnomilného druhu orchideje Gymnadenia odoratissima, na které bylo fluorescenčním mikroskopem posvíceno modrým světlem. Semínka orchidejí jsou velice malé (objekt na obrázku dosahuje kolem 1mm), a alespoň ze začátku života musí být vyživovány houbou kterou využívají. Semena jsou nabarvena propidiumiodidem.
Semínko napravo je mrtvé, což ukazují propidiumiodidem nabarvená svítící jádra (navazuje se na DNA). Druhé, stejně nabarvené, levé semínko, kterému ale jádra nesvítí, je pravděpodobně živé. Patrné je i poškození vnějšího osemení způsobeného drastickou ale nevyhnutnou sterilizací, kterou je nutno podstoupit při každém pokusu o výsev in vitro.
English: Two seeds of calciphillous orchid Gymndenia odoratissima. Seeds of orchids are very small (object on picture have around 1 mm), and young seedlings-protocorms are at early stages of ontogenesis dependent on fungi nutrition, and are parasiting on fungi. Seed on the right side is dead because of red shining nuclei (fluorescent stain propidiumiodide is bound with DNA). Seed on the left is probably alive because nuclei are not shining. Damage of the seed coat is also obvious and it was caused by drastic disinfection which is necessary if we want to germinate orchid in vitro. Fluorescent microscopy, excitation with blue light. |
